Original language description
This work is focused on development of an experimental microchip for heterogeneous immunoassay. Polystyrene substrate is used as the material for fabrication of the microfluidic chip. Techniques of hot embossing and micro-milling are employed in the fabrication. The central part of the chip contains immobilized protein molecules with biological affinity (Protein A - PA). Two procedures of PA immobilization have been studied: (i) passive sorption and (ii) covalent crosslinking of protein layers. It has been found that both these methods are proper. In the one-step assay, liquid samples containing the complementary molecules (human immunoglobulins FITC labeled - hIgG) are dosed in the place of the immobilized layer through the inlet/outlet microfluidic channels. Czech description
Tato práce je zaměřena na vývoj experimentálního čipu pro heterogenní imunoanalýzu. Polystyren byl použit jako substrát pro výrobu čipu. Při výrobě byly použity techniky mikrofrézování a slinování za tepla. Byly studovány dvě metody detekce biologickýchmolekul - lidských IgG: (i) jednokroková ProteinA-lidský IgG-FITC a (ii) sendvičová ProteinA - lidský IgG - kozí IgG-FITC. Byla vyvinuta imobiizační procedura a oběma technikami byly estrojeny kalibrační křivky závislosti fluorescenčního signálu na koncentraci IgG ve vzorku.
