The work is focused on development of microfluidic polystyrene chips for detection of biological agents. The microfluidic chips are constructed with the use of the micro-milling technology. The interaction among the immobilized bio-molecules and the complementary molecules from an analyte is the principle of the microfluidic chip. Here, the immobilized Protein A (PA) binds the complementary human immunoglobulin G (IgG) from the liquid sample and the protein-protein complex is formed. Because the immunoglobulins are fluorescently labeled, the observed fluorescence signal corresponds with the concentration of the biological macromolecule in the sample. Specificity of the interaction PA - human IgG is tested in blind experiments. Bovine serum albumin (BSA) is used as the blocking agent and Tris buffer with detergent Tween disintegrates the non-specific bonds. The experimental results demonstrate that the immunoglobulins can be quantitatively detected in the proposed multichannel microchip

Práce je zaměřena na vývoj mikrofluidních zařízení pro kvantitativní stanovení IgG. Zařízení je vyrobeno technikou mikroobrábění. Experimentální data dokazují, že kvantitativní detekce IgG v navrženém multikanálovém mikročipu je možná během několika minut.
