Chiroptical spectroscopy, a powerful tool for studying the structure, molecular geometry and conformation of chiral biomolecules, has never been systematically used for the analysis of real biofluids, except our recent studies. In our work, we address the practical aspects of measuring electronic circular dichroism (ECD), vibrational circular dichroism (VCD) and Raman optical activity (ROA) of human blood plasma, one of the most important biofluids for a sensitive screening of large populations at risk of various diseases. Human blood plasma as a complex sample often exhibits a high content of fluorescing molecules that generate a spectrally broad background of large amplitude primarily below the Raman and ROA spectra. Thus, we have developed a methodology to minimize the fluorescence effect and to improve the reliability of the ROA and Raman data. Using the combination of ROA and ECD supplemented by unpolarized infrared and Raman spectroscopies, we have monitored the spectral pattern differences reflecting conformational and stereochemical changes of chiral biomolecules that circulate in the blood plasma of patients with various diseases (cancer, metabolic, neurodegenerative). Subsequently, the spectral differences between the patients and healthy controls were evaluated by multivariate statistical methods, yielding high discrimination accuracy for example in case of pancreatic cancer. The results achieved suggest a promising perspective of chiroptical spectroscopy to reflect disease-induced pathological changes in the native states and conformations of chiral biomolecules.
