    Simple Approach for Dynamics Evaluation of Scratch Wound Healing Assay

  • Original language description

    The wound healing assay is one of the most widely used methods for studying cell migration and motility in vitro. In addition, modern automated equipment designed for time-lapse microscopy of living cells allows image sequences to be taken at short time intervals, enabling even more precise recording of cell migration dynamics. This paper describes the application of the procedure to evaluate image data acquired during wound healing of human chondrocytes and synovial sarcoma cells that were affected by the addition of synovial fluid from patients suffering from osteoarthritis. The proposed procedure is suitable for the analysis of wound healing assays even when the wound boundary cannot be detected by traditional methods, e.g. due to low cell density, numerous impurities, or when the distribution of cells forming the monolayer is affected by various factors present in the culture medium.
