    Simple Approach for Dynamics Evaluation of Scratch Wound Healing Assay

    The wound healing assay is one of the most widely used methods forstudying cell migration and motility in vitro. In addition, modern automated equipmentdesigned for time-lapse microscopy of living cells allows image sequences tobe taken at short time intervals, enabling even more precise recording of cell migrationdynamics. This paper describes the application of the procedure to evaluateimage data acquired during wound healing of human chondrocytes and synovialsarcoma cells that were affected by the addition of synovial fluid from patients sufferingfrom osteoarthritis. The proposed procedure is suitable for the analysis ofwound healing assays even when the wound boundary cannot be detected by traditionalmethods, e.g. due to low cell density, numerous impurities, or when thedistribution of cells forming the monolayer is affected by various factors present inthe culture medium.
