The 3T3 Neutral Red Uptake Phototoxicity Test (3T3NRUPT) is the only validated in vitro assay for compound phototoxicity testing that replaces the use of animals for testing cosmetics, which is banned in Europe. 3T3NRUPT uses the mouse fibroblast cell line Balb/c 3T3 (Balb/c). When the test was introduced in our studies, we dealt with a problem with Balb/c detachment from culture plates during the washing steps and following incubation in phosphate buffered saline (PBS). In order to improve the model system, we compared the attachment of Balb/c on various plate surfaces. We also tested the effect of different buffered saline solutions. In parallel, experiments were performed on a human keratinocyte cell line (HaCaT). The response of both cell lines to chlorpromazine, a recommended phototoxic compound in the validated 3T3NRUPT protocol, was also compared. Results showed a dramatic reduction in the number of Balb/c after the washing steps and subsequent incubation in buffered saline solution. In contrast, HaCaT cells were minimally affected. No beneficial effect on Balb/c number was found for any of the tested surfaces and/or buffered saline solutions. In conclusion, HaCaT cells are minimally affected during the phototoxicity assay and seems to be a suitable alternative alongside Balb/c especially for testing compounds intended to be applied topically.
