    In this study we investigated interactions of TNF-alpha and BaP in regulation of expression of CYP1B1 in rat liver 'stem-like' WB-F344 cell line. TNF-alpha enhanced induction of CYP1B1, leading to a significantly enhanced formation of DNA adducts. The increased DNA damage corresponded with potentiation of BaP-induced apoptosis induced by BaP, arrest in S-phase of cell cycle, and an increased phosphorylation of p53. Inhibition of CYP1B1 significantly decreased both formation of DNA adducts and inductionof apoptosis, suggesting that this isoenzyme might be primarily responsible for genotoxic effects of BaP in nonparenchymal liver cells.

    V této práci jsme sledovali interakci TNF-alfa a BaP a její vliv na regulaci exprese CYP1B1 u linie WB-F344. TNF-alfa zvyšuje hladinu CYP1B1, to má za následek zvýšenou tvorbu DNA aduktů a následné poškození DNA. S tím souvisí následné efekty jako je apoptóza, zástava buněk v S-fázi buněčného cyklu a fosforylace p53. Inhibicí CYP1B1 došlo ke snížení poškození DNA. Z toho vyplývá, že CYP1B1 se významně podílí na genotoxických účincích BaP.
