In this study we analyzed the molecular mechanisms of an inhibition of mast cell activity by gangliosides. Rat basophilic leukemia cells pretreated with isolated brain gangliosides exhibited decreased secretory response and calcium release. Although theproduction of phosphoinositides was also decreased, activity of phosphatidylinositol 3-kinase was not affected. Gangliosides had no effect on association of aggregated FcepsilonRI with lipid rafts and on tyrosine phosphorylation of the receptor, Syk kinase and LAT adaptor. However, they inhibited enzymatic activity of phospholipase Cgamma. This resulted in decreased production of inositol 1,4,5-trisphosphate and an inhibition of calcium response. The combined data suggest that gangliosides inhibit formation of plasma membrane-associated signaling assemblies containing phospholipase Cgamma but not early activation events.
