Ligation of TREM-1 associated with adapter DAP12 leads to tyrosine phosphorylation of the adaptor protein NTAL in a myelomonocytic cell line and primary human granulocytes. Using RNA interference to decrease the expression levels of NTAL, it was shown that in NTAL knockdown cell lines the phosphorylation of ERK1/2 is enhanced. In addition, low levels of NTAL are correlated with decreased and delayed mobilization of Ca(2+) after TREM-1 triggering. NTAL acts as a negative regulator of TNF-alpha and IL-8 production after stimulation via TREM-1. Activation signals delivered via DAP12 can be counterbalanced by the adaptor NTAL, identifying NTAL as gatekeeper of TREM-1/DAP12-induced signaling in myeloid cells.

Ligace receptoru TREM-1 asociovaného s adaptorem DAP12 vede k fosforylaci adaptorového proteinu NTAL v myelomonocytické buněčné linii a v primárních granulocytech. S využitím RNA interference k potlačení expresní úrovně NTAL bylo ukázáno, že v NTAL knockdown liniích byla zvýšena fosforylace ERK1/2. Snížená úroveň NTAL korelovala se sníženou a opožděnou mobilizací Ca(2+) po ligaci TREM-1. NTAL funguje jako negativní regulátor produkce TNF-alpha a IL-8 po stimulaci via TREM-1. Aktivační signály dodané viaTREM-1/DAP12 mohou být vyvažovány adaptorem NTAL, což identifikuje tento adaptor jako regulátor signalizace tímto receptorem v myeloidních buňkách.
