    Ubiquitin C-terminal hydrolases (UCH) are responsible for the removal of polyubiquitin chains from proteins destined for degradation in proteasomes. Thus the inhibition of deubiquitination increase the rate of proteasomal proteolysis. We detected two UCHs in the boar sperm acrosome and oocyte cortex, and shown that addition of anti-UCHs Abs increased the rate of sperm-ZP penetration and the rate of polyspermy during porcine fertilization in vitro (IVF). Sperm UCH indicating localization to acrosomal surface co-immunoprecipitated with a peripheral acrosomal membrane protein, spermadhesin AQN 1. On the other hand recombinant UCHs and isopeptidase T both reduced polyspermy when added to fertilization medium. We conclude that sperm-acrosomal UCHs are involved in sperm-ZP interaction and anti-polyspermy defense. Modulation of UCH-activity could help managing polyspermy during IVF and provide insight into male infertility.

    Ubiquitin C-terminální hydrolasy (UCH) jsou zodpovědné za odstranění polyubiquitinového řetězce z proteinů určených k degradaci v proteasomech. Inhibice deubiquitinace zvyšuje rychlost proteasomalní proteolysy. Detekovali jsme dvě UCHs v akrosomu kančí spermie a v povrchove vrstvě vajíčka, a určili, že přidáním anti-UCHs Abs se zvyšuje rychlost penetrace spermie ? ZP, a rychlost polyspermie během in vitro (IVF) fertilizace u prasat. Spermiová UCH lokalizovaná na povrchu acrosomu co-immunoprecipituje s povrchovým membránovým proteinem, AQN 1 spermadhesinem. Naopak rekombinantní UCHs a isopeptidasa T redukují polyspermii po přidání do fertilizačního media. Usuzujeme, že UCHs z akrosomu spermie se podílejí na interakci spermie-ZP a na ochraně proti polyspermii. Modulace aktivity UCH může pomoci zvládnout polyspermii během IVF a pochopit samčí infertilitu.
