Original language description
Silk protein is a biocompatible material with good mechanical properties, suitable for the development of 3D scaffolds and tissue engineering applications. The conformational transition of the secondary structure of silk fibroin due to shear stress complicates its processability using 3D printing techniques. Therefore, the solution of fibroin with polycaprolactone (PCL) in hexafluoroisopropanol as a good solvent and dimethylsulfoxid as a poor solvent were prepared. From these mixtures, 3D objects were prepared by extrusion printing and the arrangements of macromolecules in the printouts were analyzed. Using atomic force microscopy (AFM), we observed formation of polycaprolactone spherulites which were disrupted by the incorporated fibroin in the form of nano- to micro-particulate structures. The organization of the lamellae of the PCL spherulites and topography of the printed structures was rearranged by a phase separation process caused by the poor solvent in the printing dope. Resulting PCL/Fibroin scaffolds revealed good biocompatibility and mechanical stability.
