    Study of the degradome of the parasite schistosoma mansoni: role in proteolytic interactions with mammalian host

    The main objective of the project is to comprehensively characterize the spectrum of proteases identified on the basis of our transcriptomic and proteomic analyses that are actively produced by Schistosoma mansoni eggs during their interaction with host tissues, and to determine their physiological roles. The eggs represent the principal pathogenic factor of schistosomiasis, a global parasitic disease affecting more than 200 million people worldwide. They induce distinct pathological responses in different tissues, particularly the liver and intestine, while the tissue environment in turn influences the parasite itself. Two developmental stages of eggs will be investigated—immature (non-inflammatory) and mature (pro-inflammatory)—with the aim of comprehensively analyzing their excretory/secretory products, which include numerous bioactive proteins, among them proteases that remain poorly characterized. Our preliminary data reveal substantial differences in protease expression profiles between the two egg stages. By combining molecular, biochemical, and immunological approaches, the project will contribute to elucidating the mechanisms underlying host–parasite interactions and the progression of chronic disease. The project is conceived as an interdisciplinary collaboration among institutions at CZU, IOCB Prague, and UCSD, enabling efficient exchange of technologies, know-how, and students. The integrated approach includes transcriptomic data analysis, protein mass spectrometry, enzymological assays, interaction studies, and structural characterization of proteins. The outcomes will strengthen the competitiveness of the participating institutions and provide a foundation for further research activities, including funding acquisition, publication of results, and development of applied outputs.
