The impact of the gut microbiota on autoimmune mechanisms in patients with binge eating disorder and in an experimental model

Anotace anglicky

The study will follow the influence of the gut microbiota and microbial products on the onset and course of binge eating disorder (BED), the most common eating disorder. This disease is often associated with other comorbidities, such as anxiety, depression, and obsessive-compulsive disorder. In the project, we will evaluate the differences in the composition of gut microbiota and microbial metabolites in patients with BED, healthy controls with normal BMI, and obese controls to determine the extent to which the gut microbiota affects the pathophysiology of this disease. Because the gut microbiota is closely related to mucosal immunity and several inflammatory factors are produced in obese individuals, we will determine a variety of immune parameters (autoantibodies directed against appetite-regulating neuropeptides, cytokines) that may contribute to BED development. Thanks to a control group of individuals that suffer from obesity, but not BED, we will be able to distinguish the effect of obesity from the effect of binge eating. In parallel with the research of samples obtained from patients and controls, we will investigate the effect of intestinal microbiota on disease development in a mouse model of BED. This model will allow monitoring not only the parameters identical to human samples, but we will also be able to detect activity in various brain areas that are involved in pathological eating behavior. In this model, we will use the fecal microbiota transfer to assess the extent to which the intestinal microbiota is involved in the onset and course of this disease. In addition, we will monitor the effect of probiotic bacteria on the induction of BED in mice. Eating disorders and obesity can be classified as diseases of civilization and their prevalence is constantly increasing. Our project aims to contribute to the design of procedures to prevent BED and to propose a complementary nutritional therapy in the treatment of this disease.
