Biocompatible Light-Responsive Materials for Catheters, Wound Dressings and Tissue Engineering
Anotace anglicky
The long-term use of catheters and cannulas is frequently associated with inflammation and infection, posing significant risks to patients. Moreover, the increasing prevalence of multidrug-resistant pathogens and persistent infections underscores the urgent need for innovative antimicrobial strategies. This project explores the development of biocompatible light-responsive polymeric materials, including nanofiber membranes and thin polymer films, for biomedical applications such as catheters, wound dressings, and tissue engineering scaffolds. New photoactive antimicrobial materials will be developed in the form of films or nanofibers that will contain different photoactive compounds, i.e. i) porphyrinoid, ii) non-porphyrinoid type photogenerating singlet oxygen, iii) NO-photodonor, photogenerating NO radicals. The newly prepared materials will be characterized in respect to its (photo)physical and chemical properties, cell toxicity/viability with/without light irradiation of the materials, cell adhesion, proliferation, differentiation, and maturation. The effect on apoptosis, for their potential inflammatory response and hemocompatibility will be evaluated as well. In addition, their antimicrobial properties will be assessed in vitro using a variety nosocomial resistant bacterial strains (included biofilms), fungi and viruses. In-vivo application of materials modified with an antimicrobial coating to an animal model will be performed, and the tissues in contact with the materials will be analyzed histologically. By integrating advanced photodynamic inactivation mechanisms, this project aims to establish a proof of concept for biocompatible, light-responsive antimicrobial materials with spatiotemporally controlled effects. These innovations have strong potential for clinical translation, contributing to infection control in medical devices, tissue engineering, and regenerative medicine, with prospects for large-scale implementation in healthcare settings.
