Magnetic resonance (MRI) offers a wide range of images. Blood perfusion imaging (DCE) provides a powerful diagnostic tool across different medical fields. Similarly, water diffusion imaging (DWI), which is unique to MRI, delivers valuable information about tissue properties. Water diffusion can exhibit preferential directional movement, which can be quantified using diffusion tensor imaging (DTI). This anisotropic behavior is well-known within the radiological community, but recent advancements have introduced new methods capable of characterizing blood perfusion heterogeneity using contrast agents or by modifying the DTI model with tensor-based intravoxel incoherent motion (IVIM). These novel diffusion-perfusion anisotropic biomarkers may prove useful in better describing the microcapillary network of various pathologies. For example, they could enhance tumor differentiation or enable more effective and targeted radiotherapy by considering the varying oxygen effects in different tumor regions. These methods can be implemented on clinical MRI scanners; however, direct correlation with actual in vivo architecture of the microcapillary network remains impossible at present. Standard pathological techniques cannot capture the 3D heterogeneity of capillary networks, as they only provide thin 2D tissue slices. For this reason, we will investigate these MRI parameters in whole mouse tumors and correlate them with real microcapillary network images obtained via micro-computed tomography (µCT). µCT will also be used to image and quantify small, targeted samples of human brain tumors collected during MRI-guided tumor resections, allowing us to link µCT findings to their precise sampling location and correlate µCT outputs with MRI novel diffusion-perfusion parameters. This approach will demonstrate the relationship between these MRI parameters and the heterogeneity of the microcapillary network at specific tumor sites. This project partially builds upon NU21-08-00359.
