    Schizophrenia is associated with numerous abnormalities, including imbalances in all hormonal axes, among which steroids play a major role. Steroidomics studies therefore represent a promising tool for early diagnosis and appropriate treatment of schizophrenia. A group of 51 adult male schizophrenics aged 17 to 49 years were selected to investigate enzyme aberrations and the results of the circulating steroidome and other biochemical, anthropological and clinical parameters were compared with a group of 16 healthy controls aged 20 to 39 years. Both groups were tested in two phases with approximately one-year interval between the two visits. In the first step, a mixed model ANOVA including between subject factors Schizophrenia and Age group and within-subject factor Stage was used and Schizophrenia × Stage interaction was also examined. The relationships between steroids and schizophrenia were alsosimultaneously examined using multivariate regression with dimensionality reduction (Orthogonal Projection to Latent Structure, OPLS) to account for severe multicollinearity.A total of 85 analytes were monitored, including both free and conjugated steroids. Of the steroids studied, 10 correlated positively with schizophrenia, while 32 correlated negatively. Therefore, there was a significant trend towards lower steroid levels in patients with schizophrenia (p < 0.001).One year of treatment had no substantial effect on the steroidome. The level of a single steroid increased after treatment, while the levels of 8 steroids decreased and there was only borderline tendency for steroid levels to decrease after treatment (p < 0. 05). This shows that steroidomic alterations in schizophrenics are weakly treatment-dependent and primarily related to the presence of schizophrenia.
