Cryopreservation is a critical method for preserving plant genetic resources, particularly forspecies such as peach (Prunus persica), which cannot be effectively stored as seeds.However, the sensitivity of peach shoot tips to cryogenic stress often results in low viabilityand poor regrowth due to dehydration, oxidative stress, and cellular damage. This studyevaluates the potential of the surfactant Pluronic F-68 to enhance the viability and regrowthof cryopreserved shoot tips from four peach genotypes: Flame Prince, Vate 1543,Guglielmina, and Nectared 4. A droplet-vitrification approach was used, incorporatingPluronic F-68 at various stages of the process, including loading, vitrification, and post-thawrecovery solutions. Long-term exposure to PVS3 in the control group (without surfactant)significantly reduced viability, highlighting the cytotoxic effects of vitrification solutions.Compared to the control, the inclusion of Pluronic F-68 resulted in a twofold increase inshoot tip regrowth. The improved outcomes suggest that the surfactant helps stabilizemembranes, reduce osmotic stress, and mitigate dehydration damage during cryopreservation.These findings underscore the surfactant's potential as a valuable additive for optimizingcryopreservation protocols, facilitating the efficient conservation of peach germplasm forbreeding and preservation programs.
