Characterization of blaKPC-3-positive plasmids from an Enterobacter aerogenes isolated from a corvid in Canada

To examine the environmental contamination by carbapenemase-producing Enterobacteriaceae (CPE) in Canada, total of 449 faecal samples from wild corvids were analyzed. Carbapenemase production was hypothesized by MALDI-TOF MS meropenem hydrolysis assay. Carbapenemase-encoding genes were detected by PCR. Transfer experiments by conjugation were carried out with Escherichia coli recipients. blaKPC-carrying plasmids were characterized by S1 profiling and Illumina sequencing. Among 242 crbapenem-non-susceptible Gram-negative bacteria, only one KPC-3-producing Enterobacter aerogenes isolate (Eaer-4382) was identified. blaKPC-3 was transferred to E.coli laboratory strain by conjugation, while S1 analysis revealed the presence of two types of transconjugants carrying blaKPC-positive plasmids of different sizes (~90-kb [pEaer-4382s] or ~120-kb [pEaer-4382b]). pEaer-4382b (134521 bp) consisted of an IncFII-type sequence (94320 bp) and an acquired sequence (40201 bp) including a truncated KPC-3-encoding Tn4401b transposon. Plasmid pEaer-4382s (108772 bp) differed by a 25638-bp deletion of the acquired region of pEaer-4382b. Additionally, an 8-bp duplication (ACAAGATA) was identified in the region, between the resD and parA genes, resulting in a different number of iterons. Even only one KPC-3 producer was isolated from a wild corvid, the findings of the present study confirmed the environmental contamination by CPE in Canada. Furthermore, sequencing data underlined the ongoing evolution of blaKPC-carrying mobile elements.
