11th ENBDC Annual Workshop "Methods in mammary gland biology and breast cancer” Cyclin dependent kinase 12 (CDK12) is a transcription-associated kinase that when bound to cyclin K (CYCK) phosphorylates C-terminal domain of RNA polymerase II and facilitates transcription elongation. Disruption of this process results in decreased expression of DNA damage response genes, such as BRCA1, followed by genomic instability and tumorigenesis. Cdk12 knockout is embryonically lethal. Cdk12 heterozygous (het) female mice have no defects in mammary branching morphogenesis during puberty but show significant postnatal mortality of newborn pups. More than 75% of Cdk12het females failed to nurse their offspring after the first parturition. Histological analysis of the lactating mammary glands revealed accumulation of milk in Cdk12het mammary glands and qPCR showed decreased expression of milk proteins in Cdk12het lactating glands. Therefore, CDK12 might plays an important role in lactation.
