    Cytokine expression by CD163+ monocytes in Actionobactillus pleuropneumoniae infected pigs

    Actinobacillus pleuropneumoniae (APP) is a typical causative agent of lung inflammation, causing fibrino-hemorrhagic necrotizing pleuropneumonia in pigs, a disease responsible for substantial losses in the worldwide pig industry. Monocytes have an essential role in antimicrobial immune defence. During inflammation, the large subpopulation of monocytes experssing very high levels of CD163, such monocytes are called inflammatory, rapidly appear in the peripheral blood (PB), infilter inflamed tissue and increase concentration of proinflammatory cytokines such as interleukins and chemokines. The aim of the study was to evaluate contribution of CD163+ monocytes to cytokine production during APP-induced lung inflammation. Cytokines were measured by flow cytometry and by RT-qPCR in CD163+ monocytes in PB, by RT-qPCR and immunohistochemistry/immunofluorescence in lungs. Despite the systematic inflammatory reaction after APP infection, PB CD163+ monocytes did not express elevated levels of a wide range of cytokines compared to control pigs. It was shown in APP-infected pigs that, contrary to PB CD163+ monocytes, CD163+ monocytes infiltrating the lung lesion produced significant amounts of TNF-α, IL-1β, IL-6 and IL-8. In conclusion, PB CD163+ monocytes do not contribute to the elevated cytokine levels in plasma during inflammatory process. On the other hand, CD163+ monocytes contribute to proinflammatory cytokine expression at the site of inflammation during the inflammatory process.
