Aim: The aim of this study was to develop standard operating procedure SOP) for molecular detection of T. gondii oocysts in leafy green salads, implement it in several laboratories, validate it by interlaboratory analysis, and apply it in a multicentre study on ready-to-eat (RTE) salad at European level ensuring delivery of robust results. Methods: Based on the available literature, a comparative experimental work, using artificially spiked salad, was performed at two partner institutes for all the key analytical steps necessary for the SOP. The SOP was then implemented in seven consortium laboratories with the support video tutorials. A ring trial (RT) was organized to evaluate laboratory performance and efficiency. An evidence-based sampling strategy was designed to conduct a multicentre study. Conclusions: A robust and standardized procedure has been successfully implemented at European level to detect T. gondii oocyst contamination in fresh produce. Interlaboratory validation helped in defining critical steps in the procedure allowing effective corrective actions. Preliminary results of the multicentre study point on the concrete risk of RTE mixed salad as source for human toxoplasmosis.
