Host-pathogen interactions of two breeds of common carp on koi herpesvirus infection
Popis výsledku v původním jazyce
Common carp is one of the most economically valuable freshwater fish and its aquaculture importance has grown significantly during last decades. However, viral diseases can cause immense economic losses to common carp aquaculture. Koi herpesvirus (KHV) is probably the most serious viral disease affecting specifically common carp, its strains, and some hybrids with other cyprinids. Subsequently, different susceptibility of common carp strains to KHV was revealed. Current research is focused on comparing of innate immune response to KHV disease of two strains of common carp with completely opposite level of resistance to KHV: highly resistant amur wild carp (AS) and very susceptible koi carp (KOI). Objective of this study was to compare protein and gene expression profiles of two different breeds of common carp. Gene expression was analysed by qRT-PCR and protein expression profiles were analysed by mass spectrometry. Attention was paid to genes and proteins of class I interferon signalling, cell-mediated cytotoxicity and complement cascade. Although, no mortalities were observed during the experiment, KHV presence was confirmed in infected samples via quantitative RT-PCR and KHV concentration increased in during infection. Significant differences in immune response to KHV between AS and KOI were detected mostly at the level of protein expression. Gene expression analysis provided valuable new data about expression of complement cascade genes or at least confirmed previous results about expression genes related to class I signalling pathway.
