Prevention of antibiotic resistance by TARGEted Treatment of pneumonia in children
Popis výsledku v původním jazyce
The project focused on a fast, accurate and reliable method of recognizing specific pathogens, which will be an alternative to the standard identification of the causative agent of the disease using microbiological techniques or the polymerase chain reaction. A new and revolutionary method in this research is the application of the LAMP (loop-mediated isothermal amplification) method, which is able to detect certain DNA with high specificity and speed in one test tube. LAMP does not require any complex equipment or complicated detection of the resulting products. The current standard procedure for pathogen identification takes many hours to days, while the newly developed methodology reduces the entire process to minutes. This will also contribute to the fight against resistant bacteria, because we can treat patients practically immediately and we don't have to risk the so-called "blind" application of antibiotics before knowing the results of identification of pathogens using current standard methods. Such a procedure does not unnecessarily expose the patient to the side effects of ineffective antibiotics or delay the start of effective treatment, thus reducing the likelihood of the spread of resistant bacteria. Taken together, the fast and reliable LAMP method was developed for resource-limited settings to identify Streptococcus pneumoniae. We validated the method on clinical samples. Similar approach was suggested to Staphylococcus aureus, Haemophilus influenzae, and Mycoplasma pneumoniae identification.
