The acellular nerve segment is thought to be a method of choice for reparation of nerve defect. Repeated periods of freezing and thawing are frequently used for preparation of the standard acellular nerve segment in experimental application. It was believed that the basal lamina tubes and endoneurial extracellular matrix molecules survive the procedures and promote the growth of regenerating axons. We used a model of the freeze-treated nerve segment to study the molecular content of the endoneurium in absence and presence of Schwann cells. The sciatic nerves of 10 adult female rats were cut and the 10 mm long nerve segments were frozen 5 times with forceps pre-cooled in liquid nitrogen. The rats were secrificed after 14 days. Serial longitudinal frozensections, 10 ľm thick, were incubated with antibodies against S-100 protein, laminin-1, -2, and -3, fibronectin, thrombospondin, as well as chondroitin sulfate.
