The prevalence of Borrelia burgdorferi sensu lato in Ixodes ricinus ticks from South Moravia was investigated. A total of 305 ticks (21 larvae, 243 nymphs, 19 females and 22 males) were collected in suburban woods of Pisárky Park (city of Brno) from Julyto October 2002. All specimens were examined by dark-field microscopy (DFM) for the presence of spirochetes. The mean infection rate was 14.8%. 13.6% of males and 26.3% of females tested were positive for the presence of B. burgdorferi s.l.. Also, of the 243 nymphs, 14.4% were positive and out of 21 larvae 9.5% were positive. Borreliae were identified by polymerase chain reaction- restriction fragment length polymorphism (PCR-RFLP) methods. From a total of 45 DFM positive samples 37 were positive for the presence of borrelian DNA. Isolation attempts resulted in 21 spirochaetal strains.
