Hyaluronan is a native biopolymer with hydrophilic character, which contains on its chain also hydrophobic areas. Due to the prevalent hydrophilicity these parts are protected by organized water molecules. For accessing the hydrophobic parts for interactions with hydrophobic fluorescence probes the method of drying was used. During the preparation of samples for drying tert-butanol was added. The problem arise if residual part of tert-butanol remains in the sample due to its hydrophobic organic character, which can significantly affect the result of fluorescence measurement. The amount of residual tert-butanol was determined by head-space extraction with gas chromatography detection.
