Solid phase DNA extraction has been shown to be a simple method for PCR-ready DNA isolation. Poly(Llysine) coated iron oxide nanoparticles (MNPs) were not studied so far. Poly(L-lysine) coated solid surfaces are positively charged in acidic pH and negatively charged in alkali pH due to the protonation and deprotonation of functional groups. This enables the binding of charged species, including nucleic acids, using electrostatic interaction between the solid surface and the charged molecules. First, Nicotiana tabacum DNA capture and elution was studied with different amounts of poly(Llysine) coated magnetic nanoparticles. Buffers with different acidic pH (4.0 – 6.5) and alcalic pH (7.5 - 10.0) were tested for DNA capture or elution, respectively (Figure 1).
