During the last decades, it was demonstrated that polymerase chain reaction (PCR) is powerful tool of molecular biology to amplified and detect target genes and can be used for various applications such as medicine, agriculture, forensics, archaeology. The recent state-of-the-art in nanotechnology and microfabrication processes employed devices for point-of-care assays which utilize detecting of nucleic acids from ultra-low volume of sample and inexpensive screening at low cost. We demonstrate a new concept of real-time PCR and digital PCR (dPCR) in one device. For droplet real-time detection, the amplification is conducted in 0.3 µL master mixture droplet containing target gene encapsulated with 2 µL of mineral oil. For digital detection was developed chip-based system with microwell sample dispersion. The 3 µL of master mix solution with DNA is pipetted on the silicone chip and covered with a hydrophobically treated glass.
