Silk fibroin/gentamicin beads for bone regeneration
Popis výsledku v původním jazyce
Silk fibroin (SF) is a natural polymer derived from Bombyx mori and is well-studied for textile use. Last years SF attracted interest also in the field of tissue engineering due to its flexibility and biodegradability. SF can be combined synergistically with other biomaterials to create material for highly specified applications and fabricated into various forms. Incorporating antibiotics into the polymeric matrix of SF beads can prevent infection development, which is one of common post-surgery complications. SF beads with content of broad-spectrum antibiotic – gentamicin were created by gentamicine addition to the SF solution. Subsequently mixture was injected into liquid nitrogen, lyophilised and stabilised in ethanol. Cell viability was tested on human bone cell line MG63 and antibacterial activity on gram positive and gram negative bacteria. Beads morphology was examined with scanning electron microscopy. Results and discussions: Both SF beads with and without antibiotics content have similar pourous structure, which should be attractive for bone cells. Human bone cell lines treated in presence of both types of beads showed high level of viability. Inhibiotion zone analyses on agar with gram positive and gram negative bacteria proved antibacterial effect of prepared materials. Silk fibroin beads with gentamicin content showed very promising properties for bone regeneration. Antimicrobial activity of beads was proved for both gram positive and gram negative bacteria, simultaneously with the fact that gentamicin didn`t affect bone cells viability. (Poster presentation - due to COVID pandemic- virtual)
