Serial block-face scanning electron microscopy (SBEM) is a convenient method for studying the 3D structure of larger biological samples with a volume up to mm3. Ultrathin sections are cut off the specimen using an ultramicrotome placed directly inside the SEM chamber. Back-scattered electron imaging visualises the area of interest on the newly revealed sample surface. The biggest challenges we face in SBEM are sample preparation, finding the region of interest (ROI) within the sample, sample charging during the imaging, and finally, data processing and storage. Insufficient image contrast and charging often limit visualisation and make sample preparation one of the crucial steps in the SBEM analysis.
