Hop stunt viroid (HSVd) is the viroid with the broadest host range known so far, also including stone fruit trees. Until now, six viroids have been shown to be transmitted through seeds to seedlings, in one or more hosts, four of them have been detectedin pollen, cucumber strain of HSVd being among them. High incidence of HSVd infections of peach trees was recently found in the Czech Republic indicating the possibility of spreading via pollen. That is why we tested the presence of HSVd in pollen grains. Closed flowers from peach trees of cultivars Sunhaven and NJC 103 known to be infected by HSVd were collected in March. Pollen grains were separated from air dried anthers and total RNA was extracted from 10 - 20 mg of pollen or intact anthers and from100 mg of leaves or petals. PCR and molecular hybridization methods were used for HSVd detection. HSVd was consistently detected in pollen, anthers, petals as well as in leaves in each of at least three repetitions.
