Currently, deoxynivalenol (DON) and zearalenon are the only regulated Fusarium toxins in the Czech Republic. Recently, EFSA Scientific Committee proposed to extend the list of trichothecenes for which MRL is set. A novel method for determination of the trichothecenes nivalenol, deoxynivalenol, fusarenon-X, the sum of 3-acetyl-deoxynivalenol and 15-acetyldeoxynivalenol, HT-2 toxin, T-2 toxin and zearalenone in wheat has been developed using HPLC coupled with atmospheric pressure chemical ionization tandem mass spectrometry (LCMS/MS). Pressurized solvent extraction (PLE) was setted up as procedure for isolation of target compounds and its efficiency was compared with standardly used extraction (shaking with micture acetonitrile:water 84:16, v/v). Quick clean up of crude extract was performed with MycoSep# #226 or MycoSep# #225. Detection of target mycotoxins was carried out in the selected reaction monitoring (SRM) mode.
