    Evaluation of Platelet Concentrates Prepared From Whole Blood Donations With Collection Times Between 12 and 15 Minutes

    The collection time for whole blood (WB), according to Australian, Brazilian and European Guidelines, is limited to 15 minutes and donations with collection times between 12 and 15 minutes cannot be used for preparation of platelet (PLT) concentrates (PC) because of concerns about increased activation of PLT, although this is not well documented in literature. This international, multi-center study was set up to investigate the in vitro quality of PCs prepared from buffy coats (BCs) from WB with long collection times, with the aim to prevent unnecessary discarding of BCs and to simplify the total blood bank process. Each of the six centers used their standard PC preparation method and pooling system and produced 8 units from WB collections lasting < 10 min (control group) and 8 units with inclusion of one BC from a collection lasting >12 min (study group). PC were stored at 22±2 ⁰C and sampled on day 1, 6 and 8 post-collection for determination of the in vitro quality. Statistical analyses: t-tests comparing each study group with its corresponding (intra-center) control group and comparing overall groups. p<0.05 was considered significant. PC that include one buffy coat from collections lasting > 12 min have equivalent in vitro quality to controls during 8 day storage. This was confirmed by the results from one center using single-donor PC prepared with the PRP method. This study provides evidence that 12-15 minute donations should not be excluded for PC preparation, and justifies current practice in some countries.
