Single Nucleotide Polymorphisms within CCL Chemokine Genes in Pulmonary Sarcoidosis and Systemic Sclerosis

Chemokines (chemotactic cytokines) are small proteins that are secreted extracellularly and chemoattract leukocytes in a specific direction; it is along the gradient of their increasing concentration to the site of the chemokines´origin. A representativemember of CC chemokine family is MCP (monocyte chemoattractant protein) ? 1, also known as CCL2 in the systemic nomenclature, that attracts mainly mononuclear cells to site of inflammation. Other CC chemokine CCL5/RANTES (Regulated upon Activation, Normal T cell Expressed and Secreted) is a potent chemoattractant for both mononuclear cells and eosinophilic granulocytes. Because of their chemotactic activities, the up-regulation of MCP-1 and RANTES is supposed to be involved in pathogenesis of many diseases including pulmonary sarcoidosis and systemic sclerosis (scleroderma). Given a genetic component of both diseases, I postulate hypothesis that some single nucleotide polymorphisms (SNPs) within the genes encoding for MCR-1 and RANTES
