Lipopolysaccharide is a toxin released from the cell wall of Gram-negative bacteria. Infusion of lipopolysaccharide into the bovine mammary gland induces an inflammatory response [1]. Mastitis is accompanied by the production of inflammatory cytokines [2,3]. Tumor necrosis factor alpha (TNF-a) is pro-inflammatory cytokine produced mainly by macrophages and also by some of other cell types as neutrophils. The aim of this study was to analyse the effect of TNF-a on induction of lymphocyte apoptosis during inflammatory response induced by lipopolysaccharide. The experiments were carried out on the mammary glands of virgin heifers. TNF-a concentration was quantified by ELISA in four time points following stimulation of mammary glands by (24, 48, 72, 168 hours). Apoptosis of lymphocytes was analysed by flow cytometry following simultaneous staining with Annexin-V and propidium iodide in the same time points. There was found high correlation between TNF-a concentration and apoptosis of lymphocytes which suggests that this cytokine can induce lymphocyte apoptosis following influx of neutrophils caused by lipopolysaccharide.
