Searching for breed specific genetic markers of national genetic resource - the Přeštice black-pied pig

The studies of small closed pig populations play an essential role in sustainability of genetic resources. The research different molecular genetic markers is useful tool for description their breed specific and breeding management as well as keeping diversity. Especially, microsatellite sequences (Short Tandem Repeats) are used. However an analysis of other genetic markers such as Single Nucleotide Polymorphism can be helpful. The commercial multiplex PCR kit was developed for individual identification as well as kinship testing of common commercial breeds. Tetra-nucleotide microsatellite markers provided genotypes of 575 individuals of Přeštice Black-Pied Pig (PC). Special alleles - outside declared range - were detected in six markers out of eleven. Compared with other commercial breeds (LW, LA, D, PN) just in PC breed were specified three additional alleles in two markers, two new alleles in one marker and one extra allele in three markers. A hypothetical null allele in PC breed was found in one marker in 16 % all PC individuals. In this study polymorphisms in CYP2EI and MC1R genes were used to examine specific of PC breed genome. The CYP2EI gene influences skatol presence significantly. The genotype CC was proved in connection with higher amount of skatol in commercial breeds, in PC breed was not detected. The MC1R gene is consider as coat colour marker and it can be used to identification of pork products. The high frequency of Eo2 allele and the differences between PC and other breeds could be considered as primary results lead up to next research. In a wider context of protection and conservation the national genetic resources we would recommend the further research of various genetic markers across whole genome - both microsatellites and structural genes.
