Random amplified polymorphic DNA (RAPD) markers were used for the determination of the genetic stability and relationship of clones of Norway spruce (Picea abies (L.) Karst.) early somatic embryo cultures. RAPD analysis of the genetic relationship was carried out on 16 clones of somatic embryos (14 clones derived in our laboratory and two control clones derived in foreign laboratories). Twenty pre-selected primers produced 88 reproducible polymorphic and monomorphic bands, which were evaluated using theRAPDistance Program 1.04. The genetic stability during in vitro culture was studied on 6 clones derived in our laboratory (Brno, Velká Klajdovka) and two control clones. The genomic DNA was isolated in regular intervals 5 times a year during 4 subsequent years. No genetic instability was determined during the 4-year period using 4 selected primers with the highest polymorphisms.
