Covalent Antithrombin-heparin complex coated catheter prevents thrombosis in a rabbit central venous catheter model
Popis výsledku v původním jazyce
Central venous cathetrization is an essential part of modern medicine and plays a life-saving role in different groups of patients. However, obstruction due to clotting is a common problem with central venous catheters (CVCs). Surface modification of biomaterials used for manufacturing CVCs has been attempted to prevent thrombus formation. Although heparin coating is widely used for this purpose, it is far from ideal because of problems with leaching of surface-bound anticoagulant, non-uniform coating and variation in anticoagulant activities of the product. We have developed an antithrombin-heparin covalent complex (ATH) using unfractionated heparin (UFH) and antithrombin (AT). ATH has unique anticoagulant and antithrombotic properties. It has much higher anti-thrombin and anti-factor Xa activities compared to non-covalent mixtures of UFH and AT. Based on these characteristics we compared the thromboresistance of ATH-coated polyurethane (PU) catheters with uncoated or heparin-coated c
