    Diverse plasmids carrying the mcr-5 gene in Escherichia coli isolates from chicken farms in Paraguay

    Background: Recently discovered mcr genes encoding plasmid-mediated resistance to colistin have been detected globally in Enterobacteriaceae isolates from farm animals, food and human clinical samples. Several plasmid families were found to spread this novel resistance mechanism. We aim to evaluate the prevalence and characteristics of Escherichia coli isolates with plasmid-mediated resistance to colistin from broiler chicken in Paraguay and to type plasmids carrying mcr genes. Materials/methods: Cloacal samples (n=66) were taken from healthy finisher broiler chicken from 12 farms in Paraguay in 2012 and cultivated on MacConkey agar with colistin (3.5 mg/l) or cefotaxime (2 mg/l). Resistant isolates were identified by MALDI-TOF, tested for susceptibility to 13 antimicrobials and presence of plasmidmediated resistance genes mcr-1 to mcr-5. Selected E. coli isolates with mcr gene were subjected to whole genome sequencing using MiSeq Illumina platform and their phylogenic analysis was performed using CSI Phylogeny with Escherichia coli SLK172 as a reference (GenBank accession no. CP017631.1). Horizontal transfer of mcr genes was tested by conjugation experiments followed by sequencing of mcr-harbouring plasmids using MiSeq to reveal their backbone and antibiotic resistance gene content. Results: A total of 28 colistin-resistant E. coli isolates from three farms were obtained. All isolates were positive for mcr-5.1 gene, showed multi-resistance profile and diverse antibiotic resistance gene content. Most (79%) mcr-5-positive isolates produced extended-spectrum-beta-lactamase (CTX-M-8) or AmpC-type beta-lactamase (CMY-2). They carried multiple plasmid replicons (mostly I1, FII, FIB, N) and were assigned to 14 different STs with predominance of ST457 (43%). The isolates of ST457 came from all three farms and their genomes differed in 0-8 SNPs. In twenty-three (82%) isolates, conjugative transfer of mcr-5 gene into recipient strains was observed. The gene mcr-5 was carried mainly by IncI1/ST113 plasmids along with blaCTX-M-8. Several other multi-resistant mcr-5-harbouring plasmids were found including IncI1/ST12, IncHI1/ST15 and IncFII/[F29:A-:B-]/[F16:A-:B1]. Conclusions: Wide dissemination of E. coli ST457 with IncI1/ST113 plasmids carrying mcr-5 and blaCTX-M-8 was observed. Various other conjugative plasmids carrying diverse antibiotic resistance genes were associated with mcr-5. Poultry farms in Paraguay represent possible risk for public health since they may serve as a reservoir of bacteria with emerging resistance mechanisms.
