In this chapter, a more detailed influences of various biochemical factors will be added into the bone remodelling model through the RANKL-RANK-OPG chain mediating the effects of majority of acting hormones, growth factors, and cytokines. Physical activity (dynamic loading) is very essential in bone adaptation process. The addition of control factors will enable to predict concrete effects of changes of serum levels of these control factors of bone remodelling such as PTH, NO. The possibility of including vitamin D and TGF-beta into the model is also being discussed. Several diseases and their possible treatment is simulated, e.g. influence of menopause, hyperparathyroidism, and disuse.
