    Methods in molecular biology play important role in studying biological mechanisms, disease development and treatment. Therefore, learning and applying biological methods is crucial for performing experiments in research laboratories. Numerous variations in size of primary and secondary lymphoid organs and in immune responses have been described. Part of this variability is determined genetically. We studied the genetic control of weight of secondary lymphoid organs using recombinant congenic (RC) strains of mice of O20-c-B10.O20/Dem (OcB/Dem) series. Each OcB/Dem RC strain carries a unique set of 12.5% of genes (or in some cases 6.25%) from the “donor” parental strain B10.O20, and 87.5% (or in some cases 93.75%) of genes from the “background” parental strain O20. Strain OcB-43 differs from the background parental strain O20 in relative spleen and liver weight and in IgE level in serum. We analyzed genetics of these immune parameters in F2 hybrids between OcB-43 and O20. OcB-43 strain carries B10.O20-derived segments on four chromosomes. They were genotyped in the F2 hybrid mice and their linkage with immune parameters was tested by analysis of variance. We detected novel suggestive loci on chromosomes 2 and 5 influencing size of secondary lymphoid organs. Locus on chromosome 5 has a main effect on cell numbers in inguinal lymph nodes, whereas effect on relative spleen weight is observed only in interaction of loci on chromosomes 2 and 5.
