Anotace anglicky

The proposed project will focus on research of the mutual relationship between genetic and environmental risk factors; their joint impact is presumed to be the principal risk factor of the high prevalence of neurodegenerative parkinsonism in one of the regions of south-eastern Moravia, "Horňácko". The principal issues of the project will be epidemiological, genetic and environmental comparative studies in the local populations, represented by symptomatic probands and their asymptomatic relatives from the researched village Javorník and the control population. This population will be recruited in another municipality of the South Moravian region, which has no direct links to researched area and is comparable in terms of population, working habits, climate and geography. The starting point of the study are data obtained from previous epidemiological, genealogical and molecular-genetic research in the "Horňácko" region, in which multiple inter-individual presence of rare risk variants in genes associated with neurodegenerative parkinsonism was detected in symptomatic probands. We hypothesise that these variants underlie increased susceptibility to the disease, which, in combination with exposure to environmental factors, represents principal risk factor for clinical manifestation. In the framework of proposed project, the prevalence of neurodegenerative parkinsonism will be determined in the control population utilizing an epidemiological study with the same design as before in the "Horňácko" region; detailed molecular-genetic analyses will be performed in symptomatic individuals and their relatives. Environmental studies will be conducted in both populations using defined targeted and non-targeted approaches. The data obtained from the molecular-genetic and exposome analyses in both populations will then be merged and statistically analyzed, the results should answer the questions about etiology of endemic neurodegenerative parkinsonism in the "Horňácko"
