    The introduction of checkpoint inhibitors into clinical practice represented an essential milestone in the treatment of solid and some hematological malignancies. The basic principle of their effect is restoring the antitumor immune response consisting of the reactivation of T cells through inhibiting the immune checkpoints, for example, through inhibiting the interaction between PD-1/PD-L1. Thus, monoclonal antibodies such as nivolumab, pembrolizumab, cemiplimab targeted against PD-1 and atezolimumab, durvalumab, avelumab against PD-L1 were introduced into clinical practice. Although immune checkpoint inhibitors show very promising potential in treating cancer, only a part of patients respond to the treatment, and only a few achieve permanent responses. This suggests that the functions of immune checkpoints are more complex, and not yet fully described. One of the key modulators of tumor-induced suppression of the immune response is the transcription factor STAT3, which regulates PD-L1 protein expression, and whose reduced transcriptional activity leads to suppression of PD-L1 expression. This appears to be a promising approach to potentiate the effect of the therapy using monoclonal antibodies against immune checkpoints. In addition to Src inhibitors or salicylanilide derivatives, agents inhibiting the STAT3 signaling pathway can also include Janus kinase inhibitors such as JAK-STAT down-regulators. Recent first positive results of clinical trials evaluating the effect of a combination of Janus kinase inhibitors and checkpoint inhibitors showed the potential of JAK inhibitors to stimulate the function of immune cells involved in the antitumor immune response, but their effects in tumor cells remain undescribed. It is therefore still desirable to study the relationship between STAT3 signaling pathway inhibition and the interaction between PD-L1 and PD-1 in a more comprehensive manner.
