The results were presented in Agricultural and Food International Confercence, 20.-24.6.2018, in Elenite. Abstract: Authentication of animal species origin in food and feed products is important for law enforcement, and also for the protection of consumer health as well as for economic and religious reasons. Total accuracy is not always guaranteed by labelling. Therefore, an analytical approach must be used for the identification of species in processed food products. Our objective was to develop a multianalyte detection profiling technology (xMAP) based on DNA analysis for the authentication of animal species (beef, pork, chicken, horse, goat or rabbit) in food and feed products. The microsphere-based multiplex nucleic acid assay format uses different sets of magnetic microspheres in a liquid suspension as determiners of analyte specificity. In this study a specific format of multiplex oligonucleotide ligation PCR assay (MOL-PCR) was designed using the probes (oligonucleotides) targeting species-specific genetic markers (RPS7, RPS14, RPS16, mical1). The whole genome shotgun sequences for individual species were obtained from the GenBank database and were aligned for species-specific regions selection using BioEdit software. The specificity of designed oligonucleotides were checked via Blast system. DNA was isolated from muscles of each species using the DNAeasy mericon Food Kit (Qiagen) according to manual instructions. This multiplex system could be extended to detect up to 50-100 species simultaneously in one reaction and thus presents major improvement in the field of species identification in food.
