    Transmucosal drug delivery brings many advantages including delivery of macromolecules, recombinant antigens, allergens and nanoparticle-based therapeutics. The oral mucosa, especially its sublingual region, is densely populated with specialized dendritic cells and, therefore, has been recognized as a suitable immunoinductive area giving the opportunity for safe and efficient mucosal vaccination and immunomodulation. Easy access for self-administration, high permeability, a lack of enzymatic barriers, mild pH values, avoiding first-pass metabolism, all make the non-keratinized oral regions attractive sites for the application of drugs and vaccines. Many clinically used drug molecules, and also newly introduced molecules currently in various stages of development are of poor water solubility and/or low bioavailability. Due to this fact that there is a need for i) the development of new effective drug delivery systems enhancing drug bioavailability, and also ii) for the successful translation of developed delivery systems into clinical praxis. Overcoming the mucosal and skin barriers utilize various formulations strategies including nanoparticle-based delivery system, co-formulation with permeation enhancers. This is the main prerequisite for successful delivery of low-molecular drugs, immunotherapeutics and biologicals by non-invasive route of administration. Selection of proper animal models as the most important factor potecially influencing furthure results of clinical trials. In this work, we demonstrated the capability of mucoadhesive films, and transdermal delivery system to effectively deliver both, nanoparticles, and low molecules by overcoming barrier functions of skin and mucosa.
