Gallus gallus evolved to be hatched in nests and live for up to 20 years. On the other hand, broilers in commercial production are hatched in hatcheries and live for approx. 1 month. “Long-lived” layers live for approx. 1 year. Forgetting these facts results in mistaken conclusions on what is normal for poultry, including what is normal gut microbiota. Thousands of samples collected from chicken intestinal tract and environment from chicken production were collected and processed by 16S rRNA sequencing to determine microbiota composition. In next step, hundreds of pure bacterial cultures were collected from chicken intestinal tract and environment, and were used for oral inoculation of newly hatched chickens. One week later, the chicks were challenged by Salmonella Enteritidis and additional 4 days later, protective effect of administered probiotics against Salmonella infection was determined. The most efficient bacterial species were select to prepare probiotic mixtures of defined composition, tested under experimental conditions and then re-tested under field conditions at poultry farms. Bacterial species disabled in spore formation or any other way of oxygen tolerance were those that efficiently colonised chicken caecum after a single dose administration. Mixtures of such strains (Bacteroides, Megamonas, Megasphaera) increased chicken resistance to Salmonella challenge approx. 100 times. In combination with egg treatment in hatcheries, probiotic administration resulted in decrease in mortalities at commercial broiler farms from usual 4 to 3 % and allowed for antibiotic free broiler production.
