Mixed mode chromatographic columns are used for complex mixture separation. However, the ligand information of many commercially sold columns, such as the pKa of its functional group, is unknown, which complicates the method development. We propose a novel method for ionic interaction evaluation, designed for stationary phase characterisation. The method uses buffer blending by pumps to produce a range of mobile phase pH within the liquid chromatography system. The applicability of the method was utilised for ionic property elucidation of mixed mode stationary phases of weak anion-exchange character (column Acclaim WAX-1) and weak cation-exchange character (column Acclaim WCX-1). The interaction mechanism of these mixed mode columns was investigated using pump blended mobile phases in the 2.5-8.6 pH range, and observing the retention of strong acid/base analytes retention response to the dissociation/protonation of the ionic groups present on the stationary phase. This method was useful to determine the pKa value of the stationary phase ligand.
