Since ancient times, egg has been used by artists as one of the most common proteinaceous binders, because egg-based paints are very stable, dry quickly and enable painting very elaborated objects. According to written sources, the binder could consist of whole egg, only glair or yolk, respectively, or egg could be combined with additional, also non-proteinaceous materials. The type of binder influences the painting technique, i.e. paint application and layering, which is important for conservators and art historians to study artistic developments, attributions to artists or workshops, as well as deciding on conservation treatments. In this work we present a simple way for distinguishing whole egg, glair and yolk binders in cross-sections or scrapings of micro-samples from artworks by nano-LC-ESI-Q-TOF mass spectrometry. We used ratios between peptides from glair and yolk proteins, because typical glair proteins constitute major components also of yolk. The new approach was tested on 23 years old model paint layers containing yolk, glair or whole egg binders prepared as cross-sections. Besides binder identification, the influence of inorganic pigments and treatment of samples with formaldehyde on the protein analyses were studied. The method was applied to samples from five Italian artworks of the 13th, 14th and 15th centuries. The results shown in this paper can help to distinguish between different egg binders and allow identification and comprehension of so far poorly documented aspects of evolution of painting techniques.
