Lipidomic analysis is challenging due to enormous complexity of lipid structures. Two well-established analytical approaches are commonly used in lipidomics, such as shotgun ESI-MS/MS and LC/MS, both having some advantages and limitations. Alternative high-throughput and nontargeted approach is developed in this work using supercritical fluid chromatography (SFC) followed by ESI and ion mobility-mass spectrometry (IM-MS) for the determination of lipid composition. Individual parameters of SFC analysis have been carefully optimized to achieve a maximum number of separated lipid classes. Developed SFC method enables the separation of 26 nonpolar and polar lipid classes within 6 minutes. Mass spectra with high mass accuracy and resolving power are acquired using ESI in both positive- and negative-ion modes for the identification and quantitation of individual species. The quantitative analysis is performed using internal standards for each lipid class. IM is used as an additional separati
