Prunus virus F (PrVF) is new tentative species in the genus Fabavirus, first described infecting stone fruits of temperate regions. It has recently been described in the complex of viruses causing sweet cherry tree decline in the U.S.A. During a survey for Prunus spp. viruses in the Czech Republic carried out in June 2008, three asymptomatic samples of sour cherry (P. cerasus) cv. Amarelka chvalkovicka were collected (among others) in a germplasm collection in Holovousy. CLC Genomics Workbench assembly of Illumina deep sequencing reads obtained from double-stranded RNAs isolated from leaves of these trees generated 21, 25, and 12 PrVF specific contigs totaling 7.38, 5.69, and 3.48 kb for each sample, respectively. The presence of PrVF in all three trees was confirmed by RT-PCR, in both the original and newly collected samples, using primers PrVF1F (5′-TTGTTATGGGCCTCGCTCTG-3′) and PrVF1R (5′-ATCAGTTTGAAGGGAAGGCCA-3′), PrVF2F (5′-CAAGCTTTYCTTGGYGCTTT-3′) and PrVF2R (5′-GAACACGCTGGGTCATYTCA-3′) for RNA1 and RNA2 detection. Amplicons were cloned into the pGEM-T Easy plasmid and three colonies were sequenced for each of them. To our knowledge, this is the first report of PrVF infecting sour cherry in the Czech Republic. Further efforts are needed to evaluate the prevalence of PrVF in cherry populations, its mode(s) of transmission, and its potential pathogenicity.
